چکیده

Evidence is presented which establishes that chondroitin sulfate and keratan sulfate are part of the same macromolecular proteinpolysaccharide in bovine nasal septum. Degradation of the proteinpolysaccharide with papain releases single chains of chondroitin sulfate with a characteristic residual peptide still attached. A larger molecular weight fragment contains the keratan sulfate which retains a different distribution of amino acids and some chondroitin sulfate chains which can be separated by treatment with alkali with the loss of serine and threonine residues. While the ratio of protein to chondroitin sulfate varies widely in proteinpolysaccharide, the ratio of protein to keratan sulfate remains remarkably constant and different methods of degradation which release single chains of chondroitin sulfate from the macromolecule normally retain the keratan sulfate in the high molecular weight residue.

Proteinpolysaccharide complex from bovine nasal cartilage. The function of glycoprotein in the formation of aggregates.

Proteinpolysaccharide complex from bovine nasal cartilage exhibits a bimodal distribution of sedimentation coefficients when analyzed in the ultracentrifuge in associative solvents such as 0.5 M guanidinium chloride. The faster sedimenting mode disaggregates in dissociative solvents such as 4 M guanidinium chloride, giving a unimodal centrifugal pattern. Proteinpolysaccharide complex is separat...

Comparative fractionation studies of cartilage proteinpolysaccharides.

Proteinpolysaccharides from bovine embryonic rudiments and calf nasal septa were compared by conventional ultracentrifugation in isotonic salt solutions, equilibrium density gradient centrifugation, and free flow electrophoresis. These methods are all suitable for use with large amounts of material and give reliable materials balances. Fractions obtained by any one method could be isolated and ...

The Action of Hydroxylamine on the Proteinpolysaccharides of Cartilage.

In the native state, the polysaccharides of cartilage act as if they were linked to protein by bonds labile to alkali such as ester bonds. This thought was more or less clearly expressed by Krukenberg (I), Schmiedeberg (2), and Levine (3) and was based on the ease of extraction of the polysaccharides from cartilage by alkali and on their resistance to extraction by water. Practically the entire...

The separation of new forms of the proteinpolysaccharides of bovine nasal cartilage.

The principal proteinpolysaccharide, called PP-L, of bovine nasal cartilage has been fractionated centrifugally to give four products. The method requires low concentration of PP-L, high concentration of salt, and makes use of a remarkable difference between the effects of salts with monovalent and divalent cations on the sedimentation behavior of these proteinpolysaccharides. A fifth product, ...

Lanthanum Staining of Developing Chick Cartilage and Reaggregating Cartilage Cells

In this report intercellular material in developing cartilage and between reaggregating cartilage cells has been examined with lanthanum staining (3, 7, 13, 16) . The proteinpolysaccharides of cartilage have been extensively studied by biochemical methods (2, 8-10, 18-21), and proteinpolysaccharide extracted from cartilage has been shown to precipitate with lanthanum (2) . The lanthanum-stainin...
